ABSTRACT

A chemical investigation on the aerial parts of Hypericum perforatum resulted in the isolation of a new phloroglucinol derivatives (1), and seven known compounds (2-8). The structures of the compounds were elucidated by means of spectroscopic methods (MS, IR, 1D NMR, and 2D NMR) as 3-methyl-4,6-di (3- methyl-2-butenyl)-3-(4-methyl-3-pentenyl)-2-(2-ethyl-1-oxobutyl)-cyclohexanone (1),hyperforin (2),(2R,3R,4S,6)-3-methyl-4,6-di(3-methyl-2-butenyl)-2-(2-methyl-1-oxo-propyl)-3-(4-methyl-3-pentenyl)-cyclohexanone (3),hyperscabrin B (4),hyperscabrin C (5),furohyperforin isomer 1 (6),furoadhyperforin (7),and furohyperforin (8). Compound 1 was a new compound, and compounds 3-5 were obtained from H. perforatum for the first time.

ABSTRACT

Objective To discuss the feasibility of the establishment of a Y-shaped channel with windowing technique in treating the stent shunt obstruction occurring after transjugular intrahepatic portosystemic shunt (TIPS).Methods From February 2012 to December 2016,puncture windowing technique was employed in 7 patients to establish a Y-shaped channel in order to treat the stent shunt obstruction complicated by gastric varices bleeding or ascites due to recurrent portal hypertension.The preoperative Child-Pugh scores of liver function ranged from 5 to 10 points,with a mean of (6.85±1.56) points.The blood flow in both the portal vein and the shunt was determined before operation as well as at 5 days and 1,3,6months after operation;the post operative results were compared with preoperative ones.Results Y-shaped channel was successfully reconstructed in all 7 patients.The patients were followed up for a mean of 11months.Neither death nor hepatic encephalopathy occurred.Conclusion For the treatment of post-TIPS stent shunt obstruction,Y-shaped channel reconstruction within the occluded stent by using windowing technique is safe and effective,the operation is simple and easy.Therefore,this technique has certain clinical value.

ABSTRACT

Objective To study and analyze the clinical effects of rhG-CSF combined with chemotherapy in the treatment of acute leukemia. Methods 60 patients with acute leukemia were selected as the study subjects (from February 2015 to April 2017). The control group was given routine chemotherapy, the experimental group was given rhG-CSF on the basis treatment of the control group. Clinical indicators of two groups of patients were compared and analyzed. Results After the corresponding treatment, the effective rate was 63.3% in the experimental group and 43.3% in the control group, and the difference was statistically significant (P<0.05). After treatment, the quality of life score of the control group (62.30±9.20) was significantly lower than the experimental group (76.88±10.90) with statistical difference (P<0.05). The number of neutrophils reduced from 0.1×109/L to 1.5×109/L in the experimental group took (11.23±1.65), and the time for the control group was (18.90±2.78), and the difference between the two groups was statistically significant (P<0.05). There were no serious adverse reactions in the two groups, and the incidence was 10% and 13.3%, which was not statistically significant. Conclusion rhG-CSF combined with chemotherapy in the treatment of acute leukemia has good clinical effect.

Urodynamic investigation in conscious rats is widely employed to explore functional bladder disorders of various etiologies and pathogeneses. Rats can be placed in restraining cages or wide cabinets where they are allowed to move freely during cystometry. However, the requirements of special devices hampered the application of urodynamic test in freely moving rats, and whether the restraint has any effects on urodynamic parameters in conscious rats remains obscure. In the present study, we described a novel approach for urodynamic investigation in both restrained and freely moving conscious rats. In addition, we for the first time systematically compared the urodynamic parameters of rats in the two conditions. With the current method, we successfully recorded stable and repeatable intravesical pressure traces and collected expected reliable data, which supported the idea that the restraint does not affect the activity of the micturition reflex in rats, provided sufficient and appropriate measures could be applied during cystometry. Fewer technique problems were encountered during urodynamic examination in restrained rats than in freely moving ones. Taken together, conscious cystometry in rats placed in restraining cages with proper managements is a reliable and practical approach for evaluating the detrusor activity and bladder function.

ABSTRACT

Human sperm DNA is an important genetic and epigenetic material, whose chromatin structure differs from that of somatic cells. As such, conventional methods for DNA extraction of somatic cells may not be suitable for obtaining sperm DNA. In this study, we evaluated and compared three sperm DNA extraction techniques, namely, modified guanidinium thiocyanate method (method A), traditional phenol-chloroform method (method B), and TianGen kit method (method C). Spectrophotometry and agarose gel electrophoresis analyses showed that method A produced DNA with higher quantity and purity than those of methods B and C (P<0.01). PCR results revealed that method A was more reliable in amplifying DEAD-box polypeptide 4 (DDX4) and copy number variations (CNVs) than methods B and C, which generated false-positive errors. The results of sperm DNA methylation assay further indicated that methods A and B were effective, and the former yielded higher quantitative accuracy. In conclusion, the modified guanidinium thiocyanate method provided high quality and reliable results and could be an optimal technique for extracting sperm DNA for methylation assay.

ABSTRACT

ObjectiveTo evaluate the curative effect difference between Tong's anterior mesh suspension and modified tension-free vaginal tape-obturator (TVT-O). Methods75 SUI cases were random divided into two groups: Group A (35 cases) were accepted Tong's anterior mesh suspension and group B (40cases) were accepted modified tension-free vaginal tape- obturator. Curative effects, operation safety,and complications and so on, were recorded and compared. ResultsThere had no statistical differences between these two groups on operation time, bleeding lose, operation injury, immediate postoperative urinary retention andcure rates, but the operation expense of group A was cheaper than group B [ (980. 74 ±212.45)yuan vs (2879.06 ±467. 13)yuan , P <0.05). ConclusionThe curative effect between two methods were similar. It's a little complicated and fit to be popularized in large hospital to modified tensionfree vaginal tape-obturator. Tong's anterior mesh suspension was performed under completely direct vision,not through pelvic cavity, and it was an economic, convenient and easy way, which not only fit to be popularized in basic hospital, but also a surgical remedial measure to the failures who accepted these operations,such as TVT, SPARC, IVS, MONARC, TOT, TVT-O, etc.

ABSTRACT

Objective To explore the effectiveness and safety of vaginal paravaginal repair(VPVR) plus vaginal bridge repair in the treatment of female pelvic organ prolapse (POP). Methods Sixty-five patients with different defects of pelvic floor underwent VPVR or plus vaginal bridge repair for posterior vaginal wall. Patients were followed up after operation. The cure rate was estimated subjectively and objectively. The patients' quality of life was evaluated by the pelvic floor distress inventory short form 20 (PFDI-20). Results All 65 cases were treated by vaginal hysterectomy and anterior vaginal repair, in which there were 33 cases underwent VPVR while 32 cases underwent VPVR plus middle area repair. Forty concomitant procedures for vaginal bridge repair were also performed. The average operative time was (110.00±20.12) min and blood loss was (119.52±45.33) ml. The symptom of stress urinary incontinence of 25 cases significantly released after operation. Four incision recovery delayed and there were no other complicatious occurred. Patients were followed up for 6-29 months,the objective cure rate was 100.00% (65/65) and subjective cure rate was 92.31%(60/65), and 58 cases (89.23%)improved significantly with the quality of life comparing with that of pre-operation by completing PFDI-20 (P<0.01). Conclusions It is an effective and safe procedure for VPVR plus vaginal bridge repair to correct median to severe anterior vaginal prolapse and posterior vaginal wall prolapse. More clinical trials are needed to evaluate their long-term outcome.

OBJECTIVE@#To observe the changes of expression of phosphorylation c-Jun NH2-terminal kinase (p-JNK) during the skin burned wound healing in patients and discuss the molecular mechanism of burned wound healing.@*METHODS@#The staining intensity and distribution of p-JNK were detected by immunohistochemistry and routine histology in burned skin samples of 12 patients and normal skin samples of 12 control subjects.@*RESULTS@#In normal skin, the positive signals of p-JNK were mostly localized in basal layer cells of the epidermis, with a positive rate of (8.8+/-1.3)%. In the burned group, the positive signals of p-JNK were mainly localized in the epidermal cells and some inflammatory cells, with a significantly higher positive rate of (31.2+/-3.3)% than the normal group(P<0.01).@*CONCLUSION@#The changes of p-JNK expression after skin burned might correlate with wound healing.

ABSTRACT

Objective To evaluate β1、β2 and β3-adrenoreceptor mRNA expression in the sepsis rat model and to investigate the influence of the levels of β1、β2(especially β3AR)on heart dysfunction of sepsis rats.Methods The rats were randomly divided into two groups:control group and sepsis group,either group was then divided into four subgroups:6 h,18 h,24 h,36 h,RT-PCR was used to determined β1、β2 and β3-adrenoreceptor mRNA expression.Results The ratio of β1-AR/β-actin mRNA was lower in sepsis groups as compared with control.Among the two groups,the control group had the highest ratio(1.07±0.13)and sepsis 36h group had the lowest(0.59±0.13)with significant difference(P<0.05).The ratio of β2-AR/β-actin mRNA was lower in sepsis group as mmpared with coatrol,but there was no statistical significance.The ratio of β3-AR/β-actin mRNA was higher in sepsis group as compared with control.Sepsis 36 h group was the highest with an increase of 73%as compared with control(P<0.05).Conclusion These data suggest that the reduction of β1-AR mRNA and the increase of β3-AR mRNA might contribute to sepsis-related cardiac dysfunction.

ABSTRACT

<p><b>OBJECTIVE</b>To compare therapeutic effects of point sticking therapy and Chinese herbs on hyperplasia of mammary glands. Methods Sixty-three female patients with hyperplasia of mammary glands were randomly divided into an observation group (n=33) and a control group (n=30). The observation group were treated with point sticking therapy, i.e., Sanleng (Rhizoma Spargani), Ezhu (Rhizoma Zedoariae), Zhi Nanxing (Pedate Pinallia Jackinthepulpit Rhizome) and Bingpian (Borneolum) were made a plaster to apply Qihai (CV 6), Guanyuan (CV 4) and Ashi points on breast. The control group were treated with oral administration of Rupixiao.</p><p><b>RESULTS</b>After treatment of 3 courses, the total effective rate was 97.0% in the observation group, and 83.3% in the control group, the former significantly being better than the latter (P < 0.05). The score of mastalgia improvement was (13.82 +/- 5.90) points in the observation group and (7.00 +/- 3.89) points in the control group with a very significant difference between the two groups (P < 0.01).</p><p><b>CONCLUSION</b>ooint sticking therapy has a definite therapeutic effect n hyperplasia of mammary glands.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the protective effect of astragalus on testis tissues following unilateral testicular torsion/detorsion.</p><p><b>METHODS</b>Thirty healthy adult Wistar rats were equally randomized into Group A (sham-operation control), B (torsion/detorsion) and C (torsion/detorsion plus intraperitoneal injection of astragalus). The testicular torsion/detorsion model was established by the Turner method. All the rats were fed under the same condition for 7 days and sacrificed, and the torsional testes were harvested for the detection of germ cell apoptosis, glutathione activity and the level of malonic diethylaldehyde (MDA).</p><p><b>RESULTS</b>The apoptosis indexes (AI) of spermatogenic cells in the torsional testes were (5.82 +/- 1.21), (36.18 +/- 8.40) and (20.39 +/- 3.57) in Group A, B and C, significantly higher in Group B and C than in A (P < 0.05) and in Group B than in C (P < 0.05). Significant differences were observed in glutathione activity in the ipsilateral testes among Group A (48.03 +/- 2.01), B (30.93 +/- 1.25), C (38.44 +/- 1.06) U/mg (P< 0.05), as well as in the level of MDA, (1.43 +/- 0.17), (3.98 +/- 0.36), (2.57 +/- 0.53) nmol/ml, among the three groups (P < 0.05).</p><p><b>CONCLUSION</b>Astragalus could significantly reduce the apoptosis of spermatogenic cells, decrease the level of lipid peroxidation and protect glutathione activity in the torsional testis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe effects of electroacupuncture (EA) of low frequency on heroin-seeking behavior and FosB protein expression in relative brain regions so as to explore the mechanism of EA.</p><p><b>METHODS</b>Rat model of relapsing into heroin was established with progressive fixed ratio program, and model rats were randomly divided into 3 groups: a "Sanyinjiao" needle-retention control group, a low frequency and weak EA group, and a low frequency and strong EA group. Heroin-seeking behavior was elicited by conditional clue and small dose of heroin; FosB protein expression was investigated with immunohistochemical technique.</p><p><b>RESULTS</b>After treatment, the heroin-seeking behavior induced by conditional clue decreased in the needle-retention control group and the weak EA group, and the heroin-seeking behavior induced by small dose of heroin in the weak EA group significantly reduced as compared with the control group, and FosB protein expression in the nucleus accumbens septi, globus pallidus, basolateral amygdaloid nucleus significantly decreased in the weak EA group, and did not significantly change in the strong EA group; the activity induced by heroin increased as compared with those in the control group and the weak EA group.</p><p><b>CONCLUSION</b>EA of low frequency and low intensity can cure the heroin-seeking behavior, which is correlated with regulating nervous adaptation of nucleus accumbens septi, basolateral amygdaloid nucleus, etc..</p>

ABSTRACT

<p><b>OBJECTIVE</b>To examine the differences in the expression of HSP27 and c-kit between benign prostatic hyperplasia (BPH) and prostatic cancer (PCa) tissues and to analyse the relationship between their expression and BPH and PCa, especially the relationship with the occurrence, development, prognosis and treatment of PCa.</p><p><b>METHODS</b>An immunohistochemical staining (SP method) for HSP27 and c-kit was undertaken on 40 BPH and 40 PCa tissues samples.</p><p><b>RESULTS</b>Consistent patterns of cytoplasmic staining for HSP27 were seen in all sections of tissue from BPH. The glandular epithelium stained very strongly positively and the stroma stained positively. The staining for HSP27 in PCa tissues was located in the cytoplasm of glandular epithelia, but the expression of HSP27 in PCa was higher than BPH (P < 0.05). The staining for c-kit in BPH tissues was located in the cytoplasm of smooth muscle cells, and in PCa tissues was located in epithelial cells. The expression of c-kit in PCa tissues was lower than BPH (P < 0.05). The expression level of both HSP27 and c-kit were decreased with the development of grade of PCa (P < 0.05); HSP27 was increased with the development of clinical stage of PCa (P < 0.05 ); c-kit was decreased with the development of clinical stage of PCa (P < 0.05).</p><p><b>CONCLUSION</b>The expression level of HSP27 and c-kit was highly correlated with the process of the development from BPH to PCa, and also correlated with tumor grades and stages. The expression of HSP27 and c-kit may be used as an important pathological index and may be helpful for the treatment of PCa.</p>
